ISO-12-MYO1 Myograph system for isolated small vessels/ tissues
The ISO-12-MYO1 myograph system is designed for the investigation of muscular tone and its change in response to electrical, chemical or pharmacological stimuli in tissue preparations containing smooth muscle. The typical application of the ISO-12-MYO1 myograph system is the measurement of vascular tone in relation to cerebrovascular attacks, migraine, elevated blood pressure or cardiovascular side effects of new chemical entities. To answer these question needs the investigation of muscular tone of arteries, veins and small arterioles and venules even in the hundred mm diameter range in vitro. Since the myograph system detects the change in muscular tone, therefore it is capable for the investigation of any tissue preparations, which consists smooth muscle. Preparations of uterus, bladder, gall bladder, vas deferens and intestine can be studied also in isometric conditions. The sophisticated design, however, easy-to-use set-up makes it ideal for both physiological and pharmacological investigations in academic and in industrial environment as well. To avoid the corrosive effect of saline solutions the bath is coated with teflon, which provides another benefit: the chamber can be easily cleaned and residues of pharmacons can be completely removed. There are built-in light emitting diodes (LEDs) in the bottom of the chamber, which provide direct, low cost (no cold light source is needed) illumination of the bath. The chamber can be placed on a stereomicroscope stage for easier preparation. The bath is water-thermostated and is equipped with inlets and outlets for buffer superfusion. As the study requires, experiment can be done in stop-flow, continuous flow or intermittent-flow configurations. Drugs investigated can be solved in the superfused buffer or they can be added directly into the bath, which is oxygenated through a built-in pipe with a fine porous end for gentle bubbling. In case of request, platinum electrodes for electrical field stimulation can be built into the holders. Tissue fastening follows the well-established Mulvany-Halpern method, with up-to-date materials and implementation. Fixing of tungsten wires on the head-pairs is very easy with the help of spring plates. A fine micromanipulator allows the experimenter to apply pre-tension on tissue preparations. The built-in force transducer is pre-calibrated and each myograph bath comes with a calibration certificate. The transducer unit can be changed easily by the experimenter when linearity is out of range, a new unit will be provided by Experimetria Ltd. as soon as possible. The necessary data acquisition systems (hardware and software), amplifiers and circulating water bathes are also available from Experimetria Ltd, however, the force transducer provides standard data signal, therefore it can be connected any compatible electronic data acquisition system, even classical recorders. The standard MYO-01 set-up contains a bath with the following accessories: a force measuring sensor, a strain gauge amplifier with rack and power supply and a starter kit, which contains tungsten wire and a screwdriver.
